
Morpholinos屬于第三代反義寡核苷酸,由Gene Tools公司獨(dú)家研制、開發(fā)和提供合成服務(wù)。
Morpholinos主要通過阻斷蛋白質(zhì)翻譯過程而達(dá)到抑制目標(biāo)基因功能的作用,目前已經(jīng)廣泛用于發(fā)育生物學(xué)和細(xì)胞學(xué)實(shí)驗(yàn)中,并取得了非常好的實(shí)驗(yàn)效果。
Morpholinos以嗎啉環(huán)類似物為基本骨架,這種獨(dú)特的結(jié)構(gòu)特性使得Morpholinos具有以下幾個(gè)特點(diǎn):
n 與其他類型的反義寡核苷酸相比,Morpholinos能更好地抵抗核酸酶的作用
n Morpholinos與靶序列結(jié)合后通過空間位阻效應(yīng)發(fā)揮作用,不激活 RNaseH,不引起目標(biāo)基因mRNA 的降解
n 針對翻譯起始位點(diǎn)區(qū)設(shè)計(jì)的Morpholinos用于抑制蛋白質(zhì)的翻譯,針對pre-mRNA剪切位點(diǎn)區(qū)設(shè)計(jì)的Morpholinos可影響mRNA剪切,得到不同的轉(zhuǎn)錄本,可以方便地用于目標(biāo)基因結(jié)構(gòu)域的突變研究
n 與靶序列結(jié)合能力強(qiáng),可以滲入mRNA的二級結(jié)構(gòu),并且特異性好
配合Morpholinos的使用,Gene Tools公司開發(fā)了相應(yīng)的輸送系統(tǒng),可以安全、高效地將Morpholinos 輸送至胞漿內(nèi),因此,Morpholinos 也可以用于遺傳學(xué)研究和新藥靶標(biāo)的發(fā)現(xiàn)以及驗(yàn)證。此外,Morpholinos可以標(biāo)上熒光基團(tuán)、親和標(biāo)簽以及活性反應(yīng)基團(tuán),以供不同的研究需要。
